This DNT3CC monoclonal antibody reacts with mouse CCL3, also known as MIP-1 alpha (Macrophage Inflammatory Protein 1 alpha), a member of the CC chemokine subfamily closely related to CCL4 (MIP-1 beta).
CCL3 plays a critical role in leukocyte recruitment to inflammation sites, attracting monocytes, macrophages, dendritic cells, and preferentially CD8+ T cells.
It induces inflammatory cytokine secretion, mast cell degranulation, and NK cell activation, and may regulate hematopoietic stem cell quiescence.
CCL3 signals through CCR1, CCR4, and CCR5 receptors, shared with CCL4 and CCL5 (RANTES).

Applications Reported:

• Intracellular staining followed by flow cytometric analysis.

Applications Tested:

• Tested using mouse thioglycolate-elicited peritoneal macrophages with the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).
• Recommended protocol: BestProtocols® Protocol A for cytoplasmic proteins.
• Use ≤ 0.125 µg per test (100 µL final volume).
• Optimal titration recommended per assay.

Target Information

MIP-1α (CCL3) and MIP-1β (CCL4) are structurally and functionally related CC chemokines involved in immune cell trafficking and activation during infection or inflammation.
They attract macrophages, lymphocytes, NK cells, B cells, eosinophils, and dendritic cells.
MIP-1α selectively attracts CD8+ lymphocytes, while MIP-1β attracts CD4+ lymphocytes.
Both human and murine forms are active on hematopoietic cells.
