Target Information
CD73 (Ecto-5-prime-nucleotidase, 5-prime-ribonucleotide phosphohydrolase) catalyzes the conversion of purine 5'-mononucleotides to nucleosides at neutral pH, with AMP as the preferred substrate. It forms a dimer of two identical 70 kDa subunits attached externally to the plasma membrane via a glycosyl phosphatidyl inositol linkage. CD73 serves as a marker for lymphocyte differentiation, and its deficiency is associated with various immunodeficiency diseases.
Other forms of 5'-nucleotidase exist in the cytoplasm and lysosomes, distinguishable by substrate affinities, magnesium ion requirements, ATP activation, and phosphate inhibition. The CD73 gene is localized to 6q14–q21. Mutations can cause joint and arterial calcification and intestinal tuberculosis. Two transcript variants encoding different isoforms have been identified.
