Target Information

Adenosine receptors (ARs) belong to the 7-transmembrane domain G-protein-coupled receptor superfamily. Four distinct AR subtypes (A1, A2a, A2b, A3) have been identified, each mediating specific physiological processes such as cardiac rate control, smooth muscle tone regulation, platelet aggregation, inflammation, cell growth and death, and neurotransmission.

A1AR is a glycoprotein that activates Gi and Go proteins. In intact cells, agonist binding to A1AR inhibits adenylyl cyclase activity and can stimulate phospholipase C, mobilizing intracellular calcium stores. Activation of potassium channels by A1AR significantly affects cardiovascular function. A1AR is highly expressed in the brain (cerebellum, hippocampus, thalamus, cortex) and spinal cord, modulating neurotransmitter release. In white adipocytes, A1AR inhibits lipolysis and stimulates glucose uptake. It is also expressed in kidney and testis.
