Target Information
Adenosine receptors (ARs) are members of the 7-transmembrane domain G-protein-coupled receptor superfamily. Four distinct AR subtypes (A1, A2a, A2b, A3) have been identified. Activation of ARs mediates receptor subtype-specific physiological processes including cardiac rate regulation, smooth muscle tone, platelet aggregation, inflammation, cell growth, cell death, and neurotransmission.
The A1AR is a glycoprotein capable of activating Gi and Go proteins in vitro. In intact cells, agonist occupation of A1AR inhibits adenylyl cyclase activity (pertussis toxin-sensitive) and can stimulate phospholipase C, mobilizing intracellular calcium stores. Activation of potassium channels by A1AR significantly affects the cardiovascular system.
A1AR protein is highly expressed in brain regions such as cerebellum, hippocampus, thalamus, and cortex, as well as in the spinal cord, where it modulates neurotransmitter release. In white adipocytes, A1AR inhibits lipolysis and stimulates glucose uptake. Expression is also observed in kidney and testis.
