Product Specific Information

This HP-3G10 monoclonal antibody reacts with CD161 (NKR-P1A), a member of the C-type lectin superfamily. CD161 is expressed on natural killer (NK) cells and various T cell subsets, including memory/effector CD4+ and CD8+ T cells, γδ T cells, and a subset of CD3+ thymocytes. It is also found on Th17 CD4+ T cells. The receptor’s function is not fully understood, but studies suggest a stimulatory role. Lectin-like transcript-1 (LLT1), also known as osteoclast inhibitory lectin, has been identified as its ligand.

Target Information

KLRB1 (CD161) is a type II membrane protein expressed on almost all NK cells and a subset of CD3⁺ T cells. It forms a homodimeric cell surface protein (40–44 kDa) and plays an inhibitory role on NK cells. Activation of KLRB1 stimulates acid sphingomyelinase/SMPD1 and increases intracellular ceramide, as well as activating AKT1/PKB and RPS6KA1/RSK1 kinases, promoting T cell proliferation by anti-CD3.

KLRB1 binds to:

  • Gal-α(1,3)Gal and N-acetyllactosamine epitopes
  • CLEC2D/LLT1 ligand, inhibiting NK cell-mediated cytotoxicity and IFN-γ secretion

The protein contains an extracellular C-type lectin domain, a transmembrane domain, and a cytoplasmic domain.
