Product Specific Information

The monoclonal antibody OX108 recognizes human CD200R (OX2). CD200R is an inhibitory receptor structurally similar to its ligand CD200 but lacks the ITIM domain. Instead, its cytoplasmic domain contains an NPXY motif that, upon SHIP binding, inhibits ERK, JNK, and MAPK p38 pathways, leading to suppression of TNF-α secretion in activated macrophages.

CD200R expression is restricted to hematopoietic cells, including:

• Myeloid cells (monocytes, macrophages, dendritic cells, neutrophils, mast cells, basophils)
• A subset of T lymphocytes
• Langerhans cells and dendritic epidermal T cells

The epitope recognized by OX108 is thought to be near the CD200 binding site.

Applications Tested

The OX108 antibody has been tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use ≤0.5 µg/test (in 100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test. Antibody titration is recommended for optimal performance.

Target Information

CD200R (OX2R) is a receptor for the OX-2 membrane glycoprotein. Both receptor and ligand are cell surface glycoproteins containing two immunoglobulin-like domains. CD200R is mainly expressed on myeloid lineage cells and may function as a downregulatory signal. In mice, it is primarily expressed by peripheral blood monocytes and neutrophils, as well as T lymphocytes and mast cells. Alternative splicing results in multiple transcript variants. CD200R dysfunction has been associated with primary hypertrophic osteoarthropathy.
