Product Specific Information

The MEL-14 monoclonal antibody reacts with mouse CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells. It binds glycosylated, fucosylated, and sulfated glycoproteins such as CD34, GlyCAM-1, and MAdCAM-1, mediating lymphocyte rolling and homing to high endothelial venules (HEV).

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse splenocytes at ≤ 0.03 µg/test (100 µL final volume). Optimal titration recommended.

Fluorochrome Information:
Brilliant Violet™ 786 (BV786) emits at 786 nm and is compatible with cytometers equipped with a 405 nm violet laser. Ensure instrument compatibility before use.

Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
Protect vial and stained samples from light.

Fixation:
Samples may be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54). BV786 is not compatible with methanol-based fixation.

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor of the selectin family. It comprises:

• Aminoterminal C-type lectin domain
• EGF-like domain
• Two SCR sequences
• Transmembrane and cytoplasmic regions

CD62L is constitutively expressed on most leukocytes including lymphocytes, monocytes, and polymorphonuclear cells (except some memory T cells).
