Product Specific Information

The HI100 monoclonal antibody reacts with human CD45RA, a 220 kDa molecule expressed by subsets of CD4+ and CD8+ peripheral T lymphocytes and B cells. CD45RA+ T cells are primarily naive, making HI100 a useful marker for identifying T cell subsets.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-diluted and validated for flow cytometry using normal human peripheral blood cells
• Recommended Use: 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)

Brilliant Violet™ 786 (BV786) emits at 786 nm and is optimized for use with a violet (405 nm) laser. Ensure instrument compatibility before use.

Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
BV786 is sensitive to photo-induced oxidation. Protect vial and samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark. BV786 is not compatible with methanol-based fixation.

Target Information

CD45RA is an isoform of the CD45 complex with restricted expression among lymphoid cell subtypes. CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells (~10% of lymphocyte surface proteins).
It regulates Src-family kinases, influencing lymphocyte development and antigen signaling.
CD45 isoforms differ by alternative splicing and extracellular domains but share transmembrane and cytoplasmic regions.
Functions include:

• Regulation of immunoreceptor signaling
• Promotion of cell survival via integrin-mediated pathways
• Involvement in DNA fragmentation during apoptosis

CD45 is expressed on hematopoietic cells except mature red blood cells. Dysfunction of CD45 is associated with severe combined immunodeficiency and Hepatitis C.
