Product Specific Information

The GL1 monoclonal antibody reacts with mouse CD86 (B7-2), an ~80 kDa surface receptor and member of the B7 family of costimulatory molecules. CD86 is expressed at low levels on B cells, macrophages, and dendritic cells, and is upregulated upon stimulation via BCR complex, CD40, or cytokine receptors. It is also expressed by activated T cells and peritoneal cells.

CD86 interacts with T cell surface molecules CD28 and CTLA-4, playing a critical role in T-B cell crosstalk, T cell costimulation, autoantibody production, and Th2-mediated Ig production. The upregulation kinetics of CD86 suggest its major contribution during the primary immune response phase.

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of stimulated mouse splenocytes. Use ≤0.125 µg per test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best performance.

Fluorochrome Information:
Alexa Fluor 700 emits at 723 nm and is excited by the red laser (633–647 nm). Recommended filters: 685 LP mirror and 710/20 bandpass filter.

Target Information

CD86 is one of two ligands (along with CD80) for CTLA4 and CD28, acting as a costimulatory molecule in T-cell activation during antigen presentation. Insufficient co-stimulation through CD86/CD80 can induce tolerance. CD86 is a type I membrane protein of the immunoglobulin superfamily, expressed by antigen-presenting cells. It binds CD28 (activating signal) and CTLA-4 (inhibitory signal), regulating T-cell activation and immune responses. Alternative splicing produces multiple isoforms. CD86 dysfunction is associated with diseases such as gallbladder squamous cell carcinoma and myocarditis.
