Product Specific Information

The HI30 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA).
CD45 is expressed by all hematopoietic cells except circulating erythrocytes and platelets. The cytoplasmic portion has tyrosine phosphatase activity, playing a key role in lymphocyte activation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Recommended use: 5 µL (0.125 µg) per test (100 µL final volume). Cell count range: 10⁵–10⁸ cells/test.

Fluorophore Information:
APC-eFluor 780 emits at 780 nm and is excited with a 633 nm red laser. Ensure instrument compatibility.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples may be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in dark conditions.
Excitation: 633–647 nm; Emission: 780 nm; Laser: Red Laser.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, representing ~10% of lymphocyte surface proteins.
Absent in non-hematopoietic tissues, CD45 regulates Src-family kinases and cytokine receptor signaling, influencing T/B-cell activation, apoptosis, and integrin-mediated pathways.
Multiple isoforms exist due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions.
