Product Specific Information
The monoclonal antibody 20Raj1 recognizes the human Ki-67 protein, which exists in two isoforms (345 and 395 kDa) expressed in dividing cells. Ki-67 is present during active phases of the cell cycle (G1, S, G2, and mitosis) and absent in resting cells (G0).
During interphase, Ki-67 localizes to the nucleus and redistributes to chromosomes during mitosis, associating with heterochromatin-bound proteins such as CBX3.
In tumor studies, Ki-67 expression serves as a marker for determining the proliferating cell fraction.
This antibody also recognizes canine Ki-67.
Applications Reported:
- Intracellular staining using Foxp3 Staining Buffers followed by flow cytometric analysis.
Applications Tested:
- Pre-titrated and validated with Foxp3/Transcription Factor Buffers Set (Product #00-5523-00).
- Recommended use at 5 µL (0.06 µg) per test (100 µL final volume).
- Suitable for 10⁵–10⁸ cells/test (empirical determination required).
Fluorochrome Information:
- eFluor® 450 is an alternative to Pacific Blue®.

Target Information
Ki-67 is a nuclear protein expressed during cell cycle phases G1, S, G2, and M. It contains a forkhead-associated (FHA) domain and associates with euchromatin, centromeric heterochromatin, and nucleolus.
Expression pattern:
- G1: Perinucleolar
- G2: Nuclear matrix
- M phase: Chromosomal localization
Ki-67 is widely used as a proliferation marker since it is absent in G0 cells and increases from G1 through mitosis. It helps determine the growth fraction in neoplasms, correlating with tumor grade.
Also utilized as a neuronal marker for cell cycling and proliferation.
