Target Information
G protein-coupled inwardly rectifying potassium channels (Kir3.1–Kir3.4) are associated with numerous neurotransmitter receptors in the brain and are abundantly expressed in the olfactory bulb, hippocampus, neocortex, dentate gyrus, cerebellar cortex, and thalamus. Also known as GIRK or Kir3 potassium channels, they localize to the soma, dendrites, and axons of neurons.
Gβγ subunits released from G protein heterotrimers regulate Kir3 channel activity. Gβ3- and Gβ4-containing dimers enhance K⁺ current, whereas Gβ5-containing dimers inhibit it. Kir3 activity is also inhibited by tyrosine phosphorylation. Brain-derived neurotrophic factor (BDNF) activates receptor tyrosine kinase B, which phosphorylates Kir3 tyrosine residues, thereby inactivating the channels.
