Antibody detects endogenous levels of IRS1 only when phosphorylated at Tyr989.
Target Information
IRS-1 is a major substrate of the insulin receptor and becomes phosphorylated upon stimulation by insulin, IGF-1, and IL-4. Phosphorylation occurs on serine, threonine, and tyrosine residues across various tissues. Casein kinase II mediates part of the insulin-stimulated serine/threonine phosphorylation of IRS-1, with Thr502 and Ser99 identified as major phosphorylation sites in rat IRS-1. IRS-1 also contains three potential 14-3-3 binding sites (Ser270, Ser374, Ser641), with the Ser270 motif located within the phosphotyrosine binding domain responsible for insulin receptor interaction.
