Product Specific Information

The eBio500A2 monoclonal antibody reacts with the 25 kD epsilon subunit of the mouse CD3 complex.
CD3 subunits gamma, delta, and epsilon are required for proper assembly, trafficking, and surface expression of the TCR complex.
CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells, as well as NKT cells.

Binding of eBio500A2 to CD3 initiates intracellular biochemical pathways resulting in cellular activation and proliferation.
The 500A2 antibody partially cross-blocks the 17A2 and 145-2C11 antibodies, indicating overlapping epitopes.

Applications Tested: ≤0.5 µg/test (100 µL final volume; 10⁵–10⁸ cells/test)
Excitation/Emission: 633–647 nm / 723 nm (Red laser)
Filtration: 0.2 µm post-manufacturing filtered

Alexa Fluor™ 700 emits at 723 nm and can be excited with the red laser (633 nm).
Typical instrument setup: 685 LP mirror and 710/20 filter.


Target Information

The CD3 subunit complex is crucial for transducing antigen-recognition signals into T cells and regulating TCR surface expression.
It consists of CD3 gamma, delta, epsilon, and zeta subunits, encoded by closely linked genes on chromosome 11.
These subunits have long cytoplasmic tails that associate with signal transduction molecules via a double tyrosine-based motif.

CD3 is expressed on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum.
It is absent from B and NK cells.
Defects in CD3 genes are associated with CD3 immunodeficiency.
