Product Specific Information
PA3-850 detects cation-independent mannose-6-receptor (CI-MPR) in human, bovine, mouse, and non-human primate cells.
It has been successfully used in ICC/IF, Dot blot, and Western blotting procedures.
By Western blot, it detects a 300 kDa protein representing CI-MPR under non-reducing conditions.
The PA3-850 immunogen is a 300 kDa cation-independent receptor purified from adult bovine liver tissue.
Target Information
Lysosomal enzymes containing one or two mannose 6-phosphate (Man6P) moieties are transported within the cell by approximately 300 kDa cation-independent mannose 6-phosphate receptors (CI-MPR).
These receptors participate in two interconnected cycles:
- Biosynthetic cycle: Transports newly synthesized lysosomal enzymes from the trans-Golgi network to late endosomes or early lysosomes.
- Endocytic cycle: Transports extracellular lysosomal enzymes from the plasma membrane via clathrin-coated vesicles to early endosomes.
MPRs cycle between these compartments approximately every 3 hours, with the majority located in late endosomes, fewer in the trans-Golgi network, and about 10% at the cell surface.
CI-MPR also binds type II insulin-like growth factor (IGF-II), and both ligands can bind simultaneously.
A soluble truncated form (~240 kDa) representing the extracellular domain circulates in serum and retains ligand-binding capability.
