Description:
The B73.1 monoclonal antibody reacts with human CD16, a member of the Fc receptor family also known as Fc gamma RIIIa and Fc gamma RIIIb. This 50–65 kDa receptor is expressed on natural killer cells, neutrophils, monocytes, and macrophages. Crosslinking of CD16 in NK cells and monocytes activates MAPK and PI3K pathways, regulating TNF-α, IL-6, and IL-1β production.
This antibody has been shown to block Fc receptor functions.

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended ≤1 µg per test (100 µL final volume). Cell number should be empirically determined (10⁵–10⁸ cells/test). Antibody titration recommended for optimal performance.

Use in a sterile environment.

0.2 µm post-manufacturing filtered.

Target Information

CD16 encodes a receptor recognizing the Fc portion of IgG, involved in immune complex clearance and cellular cytotoxicity. The gene FCGR3A shares high similarity with FCGR3B on chromosome 1.

• FCGR3A: Expressed on NK cells, anchored via transmembrane peptide.
• FCGR3B: Expressed on neutrophils, anchored via PI linkage.

Mutations are associated with Immunodeficiency 20, Herpes Zoster, and susceptibility to Systemic Lupus Erythematosus and alloimmune neonatal neutropenia.
Alternative splicing produces different isoforms.
