Target Information
MMP9 (matrix metallopeptidase 9; also known as GELB, CLG4B) belongs to the matrix metalloproteinase family, which are zinc- and calcium-dependent endopeptidases responsible for degrading extracellular matrix proteins.
MMP9 is secreted as a 92 kDa zymogen, and upon cleavage, becomes an active 82 kDa enzyme. It features:
- A gelatin-binding domain with three fibronectin type II units
- A catalytic domain containing a zinc-binding site
- A proline-rich type V collagen-homologous domain
- A hemopexin-like domain
MMP9 is produced by monocytes, macrophages, neutrophils, keratinocytes, fibroblasts, osteoclasts, and endothelial cells, playing key roles in:
- Inflammatory responses
- Tissue remodeling and wound healing
- Tumor growth and metastasis
It degrades type IV and V collagens and is implicated in IL-8–induced mobilization of hematopoietic progenitor cells, tumor-associated tissue remodeling, idiopathic atrial fibrillation progression, and aortic aneurysm development.
