Target Information

Cas9 is an RNA-guided DNA endonuclease enzyme associated with the CRISPR (Clustered Regularly Interspaced Short Palindromic Repeats) adaptive immunity system in bacteria and genetic engineering.
This system uses RNA to direct the nuclease to specific DNA sequences, enabling:

• Indel mutations
• Sequence replacements or insertions
• Large deletions or genomic rearrangements
• Upregulation of endogenous genes
• Alteration of histone modifications or DNA methylation

Cas9 variants:

• SpCas9: Derived from Streptococcus pyogenes
• SaCas9: Derived from Staphylococcus aureus, smaller size (1053 AA vs. 1368 AA), high efficiency, nickase activity, and specificity.
  Recognizes NNGRRT PAM and performs DNA cleavage with spacer lengths of 20–24 nucleotides.
  Suitable for all-in-one delivery of Cas9 and multiple gRNA cassettes using AAV vectors.
