Product Specific Information
The RM4-5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by most thymocytes, subsets of mature T cells, and dendritic cells. CD4 binds to MHC class II molecules on antigen-presenting cells and is critical in T cell development and function. In T cells, CD4 associates with protein tyrosine kinase p56lck through its cytoplasmic tail. Binding of RM4-5 is blocked by GK1.5.
Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometry of mouse splenocytes. Recommended ≤0.25 µg/test (100 µL final volume). Adjust cell number empirically (10⁵–10⁸ cells/test). Optimize antibody titration for best performance.

Target Information
CD4 antigen is involved in recognition of MHC class II molecules and functions as a co-receptor for HIV. It is mainly expressed in T helper cells, as well as monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and certain brain and gut regions. CD4 plays a role in T-cell activation via association with the T-cell receptor complex and kinase Lck and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts are known (RefSeq, July 2017).
