Product Specific Information

The monoclonal antibody TWAJ recognizes mouse and human Gata-3, a transcription factor essential for thymic development and Th2 differentiation.
Expression is observed in the central nervous system, skin, mammary glands, and kidney during embryonic development. Homozygous knockout of Gata-3 is embryonic lethal.

Gata-3 is crucial for T cell commitment and survival, predominantly expressed in CD4 single positive thymocytes. During Th2 differentiation, Gata-3 binds to the IL-4 promoter and represses T-bet expression, inhibiting Th1 differentiation.

Alternative splice variants (50 and 45 kDa) are recognized by this antibody.
Staining requires the Foxp3/Transcription Factor Staining Buffer Set.

Applications Reported:

  • Intracellular staining followed by flow cytometric analysis.

Applications Tested:

  • Pre-titrated and tested for intracellular staining and flow cytometric analysis of mouse thymocytes using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
  • Use at 5 µL (0.125 µg) per test (100 µL final volume).
  • Suitable for 10⁵–10⁸ cells/test.

Fluorochrome Information:

  • eFluor® 660 replaces Alexa Fluor® 647.
  • Excitation: 633–647 nm; Emission: 668 nm; Laser: Red.
  • 0.2 µm post-manufacturing filtered.

Target Information

GATA3 is a zinc finger transcription factor that binds enhancer elements of T-cell receptor (TCR) genes (alpha, beta, gamma, delta).
It is expressed in T lymphocytes and regulates TCR gene activation.
The human GATA3 gene contains six exons over 17 kb DNA, with conserved zinc finger domains similar to GATA1.
No structural homologies beyond zinc fingers are observed between GATA1 and GATA3.
