Product Specific Information
- A suggested positive control for immunohistochemical applications is brain tissue.
- In Western blot, a doublet at ~65/67 kDa has been observed.
- Rabbit anti-Glutamate Decarboxylase 1/2 antibody recognizes both molecular forms of glutamate decarboxylase, GAD1 (GAD67) and GAD2 (GAD65).
- These enzymes convert glutamic acid to gamma-aminobutyric acid (GABA), the major inhibitory neurotransmitter in the brain.
Target Information
GAD-65 and GAD-67 are members of the group II decarboxylase family and catalyze the rate-limiting step in GABA production from L-glutamic acid.
- Localization:
- GAD-65: Synaptic vesicle membranes in nerve terminals
- GAD-67: Distributed throughout the cell
- Function:
- GAD-67 maintains basal GABA synthesis.
- GAD-65 activates transiently during increased neurotransmission demand.
- Physiological impact:
- Loss of GAD-65 impairs GABA neurotransmission.
- Loss of GAD-67 is lethal.
