Product Specific Information

The TMP4 monoclonal antibody reacts with human CD133 (Prominin-1), a 120 kDa member of the pentaspan family of proteins, including Prominin-2. Expression is found within plasma membrane protrusions such as epithelial microvilli. CD133 exists in alternatively spliced isoforms and has several N-linked glycosylation sites, which may vary by tissue.

Human CD133 was first identified as an epitope expressed on CD34+ hematopoietic progenitors. Although its ligand and function remain unknown, CD133 serves as a marker for stem cells and cancer stem cells. TMP4 binding does not interfere with EMK08 antibody binding, indicating distinct epitopes.

Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Use at 5 µL (0.125 µg) per test (100 µL final volume). Recommended cell count: 10⁵–10⁸ cells/test.

Target Information

CD133 (AC133) antigen is a 97 kDa glycoprotein with 5 transmembrane domains, appearing as a 120 kDa band due to glycosylation. It is a cell surface marker expressed by immature hematopoietic stem cells, not mature blood cells. CD133 also marks neural and embryonic stem cells and is expressed in certain cancers (leukemia, brain tumors). Low-level expression occurs in kidney, pancreas, placenta, and fetal liver.

Mutations in CD133 are linked to retinitis pigmentosa and Stargardt disease. Expression is regulated by at least five tissue-dependent promoters, with multiple transcript variants encoding different isoforms.
