Product Specific Information

The 1C10 monoclonal antibody reacts with mouse CD40, a 45–50 kDa type I transmembrane glycoprotein belonging to the TNFR family. CD40 is expressed by mouse B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells. It regulates B cell development and maturation by inducing Ig isotype switching and, with signals such as IL-4, promotes proliferation and prevents apoptosis.

The 1C10 clone shows agonistic activity in vitro and in vivo.

• Applications Tested: Tested on mouse splenocytes at ≤0.5 µg/test (100 µL final volume). Optimal titration recommended.
• Fluorophore Information: Super Bright 600 excited by 405 nm laser, emits at 600 nm (recommended 610/20 bandpass filter).
• Staining Recommendation: Use Super Bright Complete Staining Buffer (SB-4401) to minimize non-specific polymer interactions.
• Light Sensitivity: Protect from light due to photo-induced oxidation.
• Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (00-8222) or 1-step Fix/Lyse Solution (00-5333).
• Excitation/Emission: 405 nm / 600 nm (Violet Laser)

Target Information

CD40 is a single-chain glycoprotein and a member of the TNF/NGF receptor superfamily, homologous to CD30. It is expressed on all B cells except plasma cells, as well as some epithelial cells, carcinomas, and lymphoid dendritic cells. CD40 plays a key role in immune and inflammatory responses, including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation. Dysregulation is associated with Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency, and may be involved in Alzheimer’s disease pathogenesis.
