Product Specific Information

The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein.
CD40 is a member of the TNFR family expressed by B lymphocytes, follicular dendritic cells, thymic epithelium, and some peripheral T cells.
It regulates B cell development and maturation by inducing Ig isotype-switching and promoting proliferation in combination with IL-4.
Interaction with CD154 (gp39) on T cells is essential for T-B cell crosstalk and immune regulation.
5C3 is used for activation of B cells in in vitro functional assays.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Super Bright 600 is a tandem dye excited with the violet laser line (405 nm) and emits at 600 nm.
Recommended detection filter: 610/20 bandpass.
Ensure instrument compatibility with this fluorochrome.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity: Protect from light due to photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.

Target Information

CD40 is a single-chain glycoprotein and a member of the TNF/NGF superfamily.
It is expressed on all B cells except plasma cells, and also on epithelial cells, carcinomas, and dendritic cells.
CD40 mediates immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.
It interacts with adaptor protein TNFR2 and is implicated in amyloid-beta-induced microglial activation, suggesting a role in Alzheimer’s disease pathogenesis.
Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
CD40 dysfunction is associated with Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
