HES1 belongs to the basic helix-loop-helix (bHLH) family of transcription factors. It acts as a transcriptional repressor for genes requiring bHLH proteins for transcription. The protein contains a unique basic domain with a helix-interrupting region that binds to the N-box rather than the canonical E-box.
