Target Information

p27 Kip1 functions as a negative regulator of G1 phase progression and acts as a mediator of TGF-β–induced G1 arrest. It is a key regulator of cell cycle progression and a potent inhibitor of cyclin E- and cyclin A-CDK2 complexes. p27 Kip1 forms complexes with cyclin D-CDK4, contributing to assembly, stability, and modulation of CCND1-CDK4 activation. Its activity depends on phosphorylation state and stoichiometry, acting either as an inhibitor or activator. Degradation of p27 Kip1, triggered by CDK-dependent phosphorylation and ubiquitination by SCF complexes, is essential for transition from quiescence to proliferation. The protein has an approximate molecular weight of 27 kDa.
