Target Information
IKK alpha/beta is encoded by CHUK (conserved helix-loop-helix ubiquitous kinase) and IKKB (inhibitor of kappa light polypeptide gene enhancer in B-cells) genes located on chromosomes 10 and 8, respectively, in humans.
Upon stimulation by agents that activate the NF-κB pathway such as TNF alpha, the two subunits of the IKK complex (IKK alpha and IKK beta) alongside the regulator NEMO (IKK gamma) phosphorylate IκB proteins. This targets them for ubiquitin-mediated degradation and results in phosphorylation and nuclear translocation of NF-κB.
Phosphorylation at serine 176 and 180 in the activation loop induces a conformational change required for kinase activation.
