Target Information
STAT3는 STAT (Signal Transducers and Activators of Transcription) 단백질군에 속하며, 수용체 관련 키나아제에 의해 인산화되어 핵으로 이동 후 전사 인자로 작용합니다. CREB-binding protein과 같은 공동활성자가 STAT3의 전사 활성화에 필요합니다. STAT3는 Interferon-α, Interferon-γ, EGF, PDGF, IL6 등에 의해 활성화될 수 있으며, Tyr705에서의 JAK1 및 JAK2에 의한 인산화는 STAT3 이합체 형성, 핵 이동 및 DNA 결합 활성에 필수적입니다. 인간 STAT3 유전자는 17번 염색체 q arm에 위치하며, IFN-α에 의해 활성화되지만 IFN-β에는 반응하지 않습니다. STAT3는 세포 성장, 아폽토시스 등 다양한 세포 과정에 관여하며, Rac1 및 PIAS3와 같은 단백질에 의해 조절됩니다. 세 가지 대체 스플라이싱 변이체가 보고되었으며, STAT3 유전자 결손 마우스는 초기 배아 단계에서 치명적입니다.
