Product Specific Information

The 12G5 monoclonal antibody reacts with human CD184 (CXCR4), also termed Fusin, LESTR, or HUMSTR. CD184 is a G-protein-coupled chemokine receptor with seven membrane-spanning domains and functions as a coreceptor for X4 HIV-1 entry into CD4+ cells. It is expressed predominantly on naive T cell subsets and upregulated by PHA and IL-2 stimulation.

The 12G5 antibody partially inhibits chemotaxis and calcium influx induced by SDF-1 (the natural ligand of CD184), blocks CD4-independent HIV-2 infection, and blocks CD4-dependent infection by certain T-tropic HIV-1 isolates.

Applications Tested: Pre-titrated and tested for flow cytometric analysis of normal human peripheral blood cells. Use 5 µL (0.25 µg) per test in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.

Target Information

CXCR4 (also known as Fusin, LESTR, HUMSTR) is a member of the G-protein-coupled chemokine receptor family and serves as a co-receptor for X4 HIV-1 entry into CD4+ cells. It mediates HIV-2 binding and Env-mediated fusion, and acts as a receptor for the chemokine CXCL12/SDF-1, leading to intracellular calcium increase and MAPK1/MAPK3 activation.

CXCR4 also binds extracellular ubiquitin, influencing calcium and cAMP levels, and plays roles in hematopoiesis, cardiac and vascular development, and neuronal survival. It mediates LPS-induced inflammatory responses, including TNF secretion by monocytes. Antibodies against CXCR4 block HIV-1/2 fusion and infection of human target cells.
