Target Information
LCK는 Src-family tyrosine kinase의 일원으로, T-cell receptor (TCR) 복합체를 통한 신호전달에 필수적인 역할을 합니다.
TCR이 활성화되면 LCK는 zeta 서브유닛의 ITAM 모티프를 인산화하여 ZAP70의 SH2 도메인 결합 부위를 형성하고, ZAP70을 인산화하여 LAT 어댑터 단백질을 통한 후속 신호 플랫폼을 생성합니다.
대부분의 LCK는 세포막에 존재하지만, 일부는 Golgi apparatus와도 연관되어 있으며 이는 약한 TCR 자극 조건에서 Raf 활성화에 기여할 수 있습니다.
LCK는 myristylation 및 palmitylation 부위를 포함하고, PTK, SH2, SH3 도메인을 갖고 있어 인산화티로신 및 프롤린 풍부 모티프를 가진 단백질과 상호작용합니다.
또한 LCK는 다양한 자극에 의해 유도되는 세포사멸 조절에도 관여하나, death receptor에 의한 세포사멸에는 관여하지 않습니다.
LCK 관련 질환에는 Immunodeficiency 22 및 CD45 deficiency가 있으며, 다양한 isoform을 암호화하는 alternative splice variants가 보고되어 있습니다.
