Product Specific Information

The monoclonal antibody A1exF5 recognizes both human and mouse Arginase 1 (Arg1), a cytosolic enzyme.
This clone is compatible with standard intracellular staining protocols and the Foxp3/Transcription Factor Staining Buffer Set.

Applications Reported: Flow cytometric analysis.
Applications Tested: Flow cytometry using stimulated mouse bone marrow cells with Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).
Recommended use ≤ 0.125 µg per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test.
Careful titration recommended for optimal performance.

Fluorochrome Information:
Alexa Fluor® 700 emits at 723 nm, excited by red laser (633 nm).
Typical filter setup: 685 LP mirror and 710/20 filter.
Excitation: 633–647 nm; Emission: 723 nm; Laser: Red Laser.


Target Information

Arginase-1 (Arg1) is a 35 kDa enzyme that converts L-arginine to urea and L-ornithine, the final step in the urea cycle.
It regulates nitric oxide production by depriving NO synthase of its substrate.
Arginase 1 is expressed in:

  • Liver
  • Neutrophils
  • Myeloid-derived suppressor cells (MDSC)
  • Neural stem cells

In humans, expression occurs in blood neutrophils but not CCR3+ granulocytes.
In mice, it marks alternatively activated macrophages (M2a).
Arginase-1 is also found in myeloid cells infiltrating tumors and in hepatocellular carcinoma.
Defects cause argininemia, an autosomal recessive disorder with hyperammonemia.
