Product Specific Information
This RXDMC monoclonal antibody reacts with human and mouse Runx1 (AML1), a 50-kDa transcription factor expressed in hematopoietic cells such as myeloid, T, and B cells, but not erythroid cells. Runx1 is also detected in nearly all non-hematopoietic tissues except brain and heart.
Runx1 is involved in several stages of T cell development, including:
- CD4⁻CD8⁻ (DN) to CD4⁺CD8⁺ (DP) transition
- Commitment to the CD4 lineage
- Regulation of Th2 differentiation and immune homeostasis via interaction with Foxp3 in CD4⁺CD25⁺ regulatory T cells
- Negative regulation of Gata-3 expression
The AML1 gene encoding Runx1 is frequently translocated in acute myeloid leukemia.
Isoforms:
- Human: 11 isoforms
- Mouse: 5 isoforms (isoform 4, AML1-C, most highly expressed in hematopoietic cells)
- RXDMC antibody recognizes mouse isoform 4 and all human isoforms except AML-1FA, AML-1FB, AML-1FC.
Applications Reported:
- Intracellular staining followed by flow cytometric analysis.
Applications Tested:
- Tested by intracellular staining and flow cytometric analysis of mouse thymocytes using Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
- Recommended ≤1 µg/test (defined as staining a sample in 100 µL final volume).
- Cell number: 10⁵–10⁸ cells/test (empirical determination advised).
- Antibody titration recommended for optimal performance.

Target Information
RUNX1 is a member of the mammalian RUNX family of DNA-binding proteins regulating genes involved in cellular differentiation and cell cycle progression. It is essential for hematopoietic stem cell generation in vascular regions of the aorta, vitelline and umbilical arteries, yolk sac, and placenta. RUNX1 is expressed in all tissues except brain and heart, with highest levels in thymus, bone marrow, and peripheral blood.
