This antibody recognizes a common extracellular epitope on human HLA-DR that depends on the association of alpha and beta chains.
HLA-DR is a human MHC Class II isotype expressed on antigen-presenting cells (APCs) such as dendritic cells, B lymphocytes, monocytes, and macrophages.

Target Information

HLA-DR is a transmembrane glycoprotein composed of a 36 kDa alpha chain (DRA) and a 27 kDa beta chain (DRB).
The DRA gene contains 5 exons:

• Exon 1: leader peptide
• Exons 2–3: extracellular domains
• Exon 4: transmembrane and cytoplasmic regions

DRA acts as the sole alpha chain for DRB1, DRB3, DRB4, and DRB5, while the DRB chain carries polymorphisms defining peptide-binding specificity.
Hundreds of DRB1 alleles have been identified, and HLA-DR typing is routinely performed for bone marrow and kidney transplantation.

HLA-DR is primarily expressed on:

• B lymphocytes
• Monocytes
• Macrophages
• Thymic epithelial cells
• Activated T lymphocytes
