Product Specific Information

• Source: Monoclonal antibody obtained by fusing the P3-X63-Ag8 mouse myeloma cell line with spleen cells of Donryu rat after immunization with mouse teratocarcinoma and mouse liver E-cadherin fragment.
• Specificity: Specifically reacts with mouse E-cadherin. Does not inhibit E-cadherin-dependent cell-cell contact.
• Cross Reactivity: Reacts with human and dog E-cadherin. Does not react with bovine or chicken E-cadherin.
• Western Blot: Can be performed under reducing or non-reducing conditions.

Reconstitution and Dilution

• Dissolve the lyophilized antibody in 50 µL of distilled water to obtain a final concentration of 2.0 mg/mL (stock solution).
• For dilution, use 20 mM TBS (pH 7.5), 10 mM CaCl₂, 1.0% BSA, 0.1% NaN₃.
• Note: Use TBS for dilution; PBS causes precipitation.

Storage

• Add 0.1% sodium azide as a preservative when storing at 4ºC.
• Store antibody at ≥2.0 mg/mL for stability.
• Stock solution (2.0 mg/mL):
  • Store at -20ºC for 1 year, or
  • Store at 4ºC for 6 months (with 0.1% sodium azide).
• Avoid repeated freeze-thaw cycles.
• Do not store diluted antibody.

Target Information

E-Cadherin (epithelial cadherin) is a calcium-dependent cell-cell adhesion glycoprotein from the cadherin superfamily. It consists of five extracellular cadherin repeats, a transmembrane region, and a conserved cytoplasmic tail.
Mutations in the CDH1 gene (chromosome 16) are associated with various cancers including gastric, breast, colorectal, thyroid, and ovarian cancers.
Loss of E-cadherin function contributes to cancer progression through increased proliferation, invasion, and metastasis.
E-cadherin also mediates bacterial adhesion and is essential for tissue architecture and integrity.
