• Heat-mediated antigen retrieval is recommended prior to staining using 10 mM citrate buffer (pH 6.0) for 10–20 minutes, followed by cooling at room temperature for 20 minutes.
• After retrieval, incubate samples with primary antibody for 10 minutes at room temperature.
• Suggested positive control: tonsil tissue.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin family that plays an integral role in signal transduction and T cell differentiation and activation.
CD8 is predominantly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor with the T cell receptor (TCR) for MHC class I molecules.
CD8 also exists as homodimers of CD8α on subsets of T cells, NK cells, monocytes, and dendritic cells.
Upon MHC-I/peptide complex ligation, CD8 recruits Lck kinase, leading to cytokine production, motility, and cytotoxic T lymphocyte (CTL) activation.
Activated CTLs are critical for pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
