Target Information
Amyloid beta peptide (Abeta/Beta-amyloid)은 Alzheimer’s disease 환자의 뇌에서 발견되는 amyloid plaque의 주요 구성 성분입니다.
이 펩타이드는 40–43개의 아미노산으로 구성되어 있으며, beta-amyloid precursor protein (beta APP)으로부터 두 단계의 절단 과정을 통해 생성됩니다.
- 첫 번째 단계: beta APP의 아미노 말단 도메인이 aspartyl protease인 beta-secretase (BACE)에 의해 절단됩니다.
- 두 번째 단계: 잔여 막 결합 단백질이 gamma-secretase에 의해 절단되어 amyloid beta peptide가 형성됩니다.
Gamma-secretase는 presenilin-1, presenilin-2, nicastrin 등의 단백질로 구성된 복합체로 알려져 있습니다.
Abeta는 amyloid cores, neuritic plaques, neurofibrillary tangles 등에 존재하며, Alzheimer’s disease, Lewy body dementia, Down’s syndrome, amyloidosis (Dutch type), cerebroarterial amyloidosis, Guam Parkinson-Dementia complex 등과 관련이 있습니다.
