Target Information
This gene encodes a protein that plays a major role in the bidirectional stimulation of T and B cells. The protein contains an SH2 domain and a short tail, and associates with the signaling lymphocyte-activation molecule (SLAM), acting as an inhibitor by blocking recruitment of SHP-2 to its docking site. It also binds other related surface molecules expressed on activated T, B, and NK cells, modulating signal transduction pathways. Mutations in this gene cause X-linked lymphoproliferative syndrome type 1 (Duncan disease), a rare immunodeficiency characterized by extreme susceptibility to Epstein-Barr virus infection, severe mononucleosis, and malignant lymphoma. Multiple transcript variants encoding different isoforms have been identified.
