Specificity: Antibody detects endogenous levels of QIK/QSK only when phosphorylated at Thr182/Thr175/Thr221.
Target Information
Serine/threonine-protein kinase (SIK1) is involved in multiple cellular processes including cell cycle regulation, gluconeogenesis, lipogenesis, muscle differentiation, and tumor suppression. It phosphorylates targets such as HDAC4, HDAC5, PPME1, SREBF1, and CRTC1/TORC1, thereby modulating CREB activity and metabolic pathways.
SIK1 acts as a tumor suppressor via p53-dependent anoikis and regulates sodium signaling through phosphorylation of PPME1 and ATP1A1. It also influences muscle gene expression and cardiomyogenesis by controlling HDAC4/5 and CDKN1C/p57Kip2.
SIK2 modulates insulin signaling by phosphorylating IRS1 (Ser-794).
SIK3 regulates mTOR signaling through DEPTOR degradation and contributes to chondrocyte differentiation during skeletogenesis. It also negatively regulates cAMP signaling by acting on CRTC2/TORC2 and CRTC3/TORC3, and prevents HDAC4 nuclear translocation.
