Target Information
The tumor necrosis factor (TNF) receptor superfamily consists of several type I integral membrane glycoproteins sharing homology in their cysteine-rich extracellular domains. Members include TNF-RI, TNF-RII, and CD40. Ligands for these receptors can be small, secreted proteins such as TNF, or type II membrane proteins like CD40L.
Activation of TNF-R or CD40 induces nuclear translocation of NFκB, with TRAF (TNF receptor-associated factor) family members implicated in this process. TRAF4, originally termed CART1, is specifically expressed in breast carcinomas and localized to the nucleus in such tissues.
