Product Specific Information
The eBioGHI/61 monoclonal antibody reacts with human CD163 (Haemoglobin Scavenger Receptor, M130), a member of the macrophage scavenger receptor family.
CD163 is expressed by tissue macrophages, circulating blood monocytes, and possibly by hematopoietic progenitor cells. It functions as a high-affinity receptor for the hemoglobin-haptoglobin complex to clear hemoglobin from blood. Ligand binding triggers signal transduction leading to IL-6 and IL-10 production.
CD163 expression is induced by IL-6 and IL-10, while proinflammatory stimuli like LPS or TNF-α cause shedding of soluble CD163.

Target Information
CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein and receptor for the hemoglobin-haptoglobin complex. It protects tissues from oxidative damage and plays a role in iron recycling via endocytosis.
CD163 is expressed exclusively on human monocytes and macrophages, predominantly in late inflammation. It is present on all CD14⁺ monocytes and most CD64⁺ monocytes, showing higher expression on CD16⁺ subsets.
Expression is upregulated by IL-10, IL-6, dexamethasone, and shed by LPS or PMA stimulation. CD163 binding is calcium- and pH-dependent, showing higher affinity for multimeric haptoglobin (HP1F phenotype).
Intracellular signaling involves tyrosine kinase-dependent calcium mobilization, inositol triphosphate production, and secretion of IL-6 and CSF1.
