Product Specific Information

The RA3-6B2 monoclonal antibody reacts with the exon A-restricted isoform of mouse CD45, a 220 kDa surface molecule. CD45R/B220 epitope is mainly expressed by B cell lineage from early Pro-B to mature B cells. Some activated T cells, lymphokine-activated killer (LAK) cells, NK cell progenitors, and certain mutant T cells (lpr/lpr) also express this antigen.

Applications Reported:
Flow cytometry and immunohistochemistry (frozen tissue sections).

Applications Tested:

• Flow cytometry: ≤0.25 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
• IHC (Frozen): ≤10 µg/mL.
  Careful titration recommended for optimal performance.

Target Information

CD45R (PTPRC) belongs to the protein tyrosine phosphatase (PTP) family, regulating cell growth, differentiation, and signaling. CD45 is expressed in hematopoietic cells and is essential in T- and B-cell receptor signaling. It modulates Src-family kinases and integrin-mediated pathways, influencing lymphocyte development and apoptosis. Multiple isoforms exist due to alternative splicing, differing in extracellular domains. CD45 dysfunction is linked to Severe Combined Immunodeficiency and Hepatitis C.
