Target Information
Spleen tyrosine kinase (SYK), along with ZAP70, are cytoplasmic tyrosine kinases considered potential drug targets for allergic disorders and autoimmune diseases. SYK may also play a role in cancer. Upon B cell antigen receptor (BCR) or high-affinity IgE receptor (FcepsilonRI) activation, Syk translocates to the plasma membrane and phosphorylates downstream adaptor proteins, initiating signaling pathways such as calcium mobilization, cytoskeleton remodeling, or transcription of specific genes. Syk interacts with receptor assemblies via its SH2 domains binding to ITAM motifs phosphorylated by Src-family kinases, which also activate Syk by phosphorylation of its activation loop.
