Target Information
The structure-specific ERCC1/XPF endonuclease complex is involved in the repair of two distinct types of DNA lesions:
- Nucleotide excision repair (NER) for UV-induced lesions and bulky chemical adducts
- Recombination repair of genotoxic interstrand cross-links
In NER, dual incisions occur on both sides of the DNA damage. In mammalian cells, XPG cleaves 3′ of the lesion, while the ERCC1/XPF complex makes the 5′ incision. ERCC1/XPF also likely participates in processing partially repaired recombination intermediates.
