Product Specific Information
Store vial at -20°C prior to opening. Aliquot contents and freeze at -20°C or below for extended storage. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4°C as an undiluted liquid. Dilute only prior to immediate use.
Anti-p27 Kip1 pS140 is directed against the phosphorylated form of human p27 Kip1 protein at the S140 residue. This antibody specifically reacts with p27 Kip1 protein from human tissue. Reactivity against p27 from other species is unknown.
Target Information
p27 Kip1 functions as a negative regulator of G1 phase progression and is a possible mediator of TGF-β induced G1 arrest. It is an important regulator of cell cycle progression and a potent inhibitor of cyclin E- and cyclin A-CDK2 complexes. p27 Kip1 forms complexes with cyclin D-CDK4 and influences assembly, stability, and activation of CCND1-CDK4 complexes. Depending on phosphorylation state and stoichiometry, p27 Kip1 can act as either an inhibitor or activator of cyclin D-CDK4 complexes. It has a molecular weight of approximately 27 kDa, and its degradation—triggered by CDK-dependent phosphorylation and subsequent ubiquitination by SCF complexes—is essential for cellular transition from quiescence to proliferation.
