Target Information
PKN2 (PRK2), a close relative of the PKCs, is a regulator of mitosis and exit from cytokinesis. It plays roles in cell cycle regulation, actin cytoskeleton assembly, cell migration, adhesion, tumor invasion, and transcriptional activation signaling.
It phosphorylates CTTN in hyaluronan-induced astrocytes, reducing its association with filamentous actin, and phosphorylates HDAC5, impairing its nuclear import.
PKN2 acts as a direct RhoA target, required for apical junction formation in epithelial cells, and is essential for G2/M phase progression and cytokinesis in an ECT2-dependent manner. It stimulates FYN kinase activity during keratinocyte differentiation and regulates epithelial bladder cell migration.
Additionally, PKN2 inhibits Akt pro-survival kinase activity and mediates Rho-induced transcriptional activation through the c-fos SRF pathway. It also phosphorylates HCV NS5B, stimulating HCV RNA replication.
