Product Specific Information
The AA4.1 monoclonal antibody reacts with mouse C1qRp, a 130–140 kDa C-type lectin-like type I transmembrane protein.
Originally identified as an antigen expressed during early B cell development in bone marrow, AA4.1-positive cells are also found in adult spleen and fetal liver.
In combination with markers such as Thy-1, Sca-1, c-Kit, CD43, and CD24, AA4.1 expression helps distinguish primitive and differentiated subsets of lymphoid-committed progenitors (CLP).
Surface expression of IgM, CD23, and AA4.1 defines three transitional B cell subsets:
- T1: AA4.1+/CD23-/sIgM^hi
- T2: AA4.1+/CD23+/sIgM^hi
- T3: AA4.1+/CD23+/sIgM^lo
AA4.1 is also expressed in endothelial cells, megakaryoblasts, and platelets.
Monoclonal antibodies AA4.1 and 493 recognize different epitopes of the same molecule.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse bone marrow and splenocyte cells at ≤0.5 µg/test (in 100 µL). Cell count: 10⁵–10⁸ cells/test.
Optimize titration empirically for best performance.
Light Sensitivity: Protect from light; tandem dye is photo-oxidation sensitive.
Fixation: Samples can be stored up to 3 days at 4°C in the dark using:
- IC Fixation Buffer (Product #00-8222-49)
- 1-step Fix/Lyse Solution (Product #00-5333-54)
Excitation: 488–561 nm

Target Information
The encoded protein is a cell-surface glycoprotein and type I membrane protein, initially identified as a myeloid cell-specific marker.
Although once thought to be a C1q receptor, it is now associated with intercellular adhesion and clearance of apoptotic cells.
Its cytoplasmic tail interacts with moesin, linking transmembrane proteins to the cytoskeleton and contributing to cytoskeletal remodeling.
