Product Specific Information

The monoclonal antibody IL22JOP reacts with and inhibits the bioactivity of human and mouse IL-22. IL-22 is a 20 kDa member of the IL-10 cytokine family, secreted primarily by Th17 cells, NK cells, and other T cells. Compared to IL-6 or TGF-beta, IL-23 can induce greater levels of IL-22 in in vitro-differentiated Th17 cells, suggesting IL-22 secretion from more fully differentiated Th17 cells in vivo.

IL-22 has been shown to protect hosts from bacterial infections of the lungs and gut. Elevated IL-22 production has been observed in PBMCs from asthma patients, and increased IL-17⁺IL-22⁺ CD4 T cells have been found in ankylosing spondylitis and rheumatoid arthritis patients.

IL22JOP is reported to recognize rhesus IL-22.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested on Th17-polarized CD4⁺ normal human peripheral blood cells. Recommended ≤0.25 µg/test (100 µL final volume). Cell numbers may range from 10⁵ to 10⁸ cells/test.
Excitation: 633–647 nm

Target Information

IL-22, also known as IL-10-related T-cell derived inducible factor, is an alpha-helical cytokine and a member of the IFN–IL-10 family (including IL-19, IL-20, IL-24, IL-26, IL-28, IL-29, and type I/II interferons). It is produced mainly by activated T cells and NK cells. In humans, the IL-22 gene is located on chromosome 12q and is structurally related to IL-10.

IL-22 acts via a heterodimeric receptor complex composed of IL-22R1 and IL-10R2, activating the JAK-STAT signaling pathway. A soluble receptor, IL-22BP, antagonizes IL-22 binding and signaling. IL-22 primarily targets nonimmune cells such as epithelial cells, hepatocytes, and keratinocytes, influencing innate immunity, wound healing, and inflammation.

IL-22 is co-expressed with IL-17A and IL-17F by Th17 cells but is differentially regulated. It induces acute phase reactants, antimicrobial proteins, and keratinocyte mobility. IL-22 is upregulated during chronic inflammation and plays a critical role in defense against extracellular pathogens and tissue repair.
