Target Information
EIF2AK2 (PKR) is one of four kinases that specifically phosphorylate Ser51 of translation initiation factor eIF2-alpha in response to environmental stresses. It plays a key role in antiviral defense through activation by double-stranded RNA produced during viral infection. PKR mediates IFN-induced antiviral responses, virus-induced apoptosis, and regulates cell growth and differentiation. It activates Fas expression and FADD/Caspase-8 signaling, functioning as a dual protein that sequentially activates both survival and death pathways. PKR is involved in NF-kappaB, p53, p38, and PDGF signaling, and has implications in tumor suppression and neurodegenerative diseases such as Alzheimer’s, Huntington’s, and ALS. Viruses can inhibit PKR activity, e.g., influenza virus NS1 protein.
