Description:
The eBioCB16 monoclonal antibody recognizes CD16 (Fc gammaRIII), a low-affinity receptor for IgG with an apparent molecular weight of 50–80 kDa. CD16 includes two similar genes, CD16A (FcγRIIIA) and CD16B (FcγRIIIB).

  • CD16A: expressed in macrophages and NK cells (polypeptide-anchored form)
  • CD16B: expressed in neutrophils (GPI-anchored form)

Two polymorphisms, NA-1 and NA-2, are known for CD16B. NA-2 homozygotes show reduced phagocytic capacity compared with NA-1. CD16 binds IgG immune complexes, showing preferential binding to IgG1 and IgG3, minimal to IgG2 and IgG4. Upon IgG binding, CD16 initiates signaling cascades leading to ADCC, phagocytosis, degranulation, and proliferation.

Applications Reported:
This antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Validated by flow cytometry on normal human peripheral blood cells. Recommended usage ≤ 0.125 µg per test (100 µL final volume). Optimal titration should be determined experimentally.


Target Information

CD16 encodes a receptor that recognizes the Fc portion of IgG and participates in immune complex clearance, cellular-mediated cytotoxicity, and viral infection enhancement.

  • Related gene: FCGR3B (similar sequence, expressed in PMN)
  • FCGR3A: membrane glycoprotein anchored via transmembrane peptide
  • FCGR3B: anchored via phosphatidylinositol linkage

Mutations in FCGR3A are associated with immunodeficiency 20, recurrent viral infections, systemic lupus erythematosus, and alloimmune neonatal neutropenia. Alternative splicing produces different isoforms.
