Product Specific Information
Store vial at 4°C prior to restoration. For extended storage, aliquot contents and freeze at -20°C or below. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4°C as an undiluted liquid. Dilute only prior to immediate use.
Acid Phosphatase is an IgG fraction antibody purified from monospecific antiserum by a multi-step process including delipidation, salt fractionation, and ion exchange chromatography, followed by extensive dialysis against the buffer stated above. Immunoelectrophoresis shows a single precipitin arc against anti-Biotin, anti-Rabbit Serum, as well as purified and partially purified Acid Phosphatase [Potato]. Cross-reactivity against Acid Phosphatase from other sources is unknown.
Target Information
Low molecular weight protein tyrosine phosphatases, including the human red cell phosphatase HCPTP, are widely expressed cytosolic proteins (~18 kDa) existing in distinct isoforms. These enzymes are highly selective for phosphotyrosine over phosphoserine or phosphothreonine. HCPTP-A and HCPTP-B, expressed in human red cells and placenta, correspond to fast and slow forms of red cell acid phosphatase. Like other tyrosine phosphatases, HCPTP is inhibited by vanadate and utilizes a catalytic mechanism involving a cystinylphosphate intermediate. The crystal structure of bovine heart-derived enzyme BHPTP reveals a four-stranded central parallel β-sheet with flanking α-helices and an active site cysteine residue in the typical tyrosine phosphatase sequence motif CXXXXXR.
